I assume that you are referring to Ephesians 5:27: "That he might present it to himself a glorious church, not having spot, or wrinkle, or any such thing; but that it should be holy and without blemish."
The phrase "not having spot or wrinkle" equals to the other phrase "without blemish." These blemishes, such as spots or wrinkles, refer to sin or secret faults.
I like to use the analogy of a white shirt. In order for the white to be perfect, there can be no stains, no spots, no wrinkles. If there is ANY kind of blemish, the shirt is no longer perfect.
God's desire for the church is to be spotless and wrinkle-less of sin!
